Plant-Pathogen Interactions Sharma, Anubhuti [1], Sharma, Vinay [2]. Host-Pathogen Interaction in Sesamum indicum. Sesamum (Sesamum indicum L.), an important oil yielding crop of the semi-arid region contains high amount of oil (50%) and protein (18-20%) in its seeds. However sesamum is susceptible to infection by several pathogens that causes huge economic losses. Tiarosporella phaseolina is a major pathogen of this species causing ‘root rot’ disease which results in heavy losses to the crop. The present study was undertaken to know the biochemical basis of disease resistance of the rot disease with particular reference to phenolics. The healthy leaves of different ages (7 days old and 15 days old plants) were inoculated with the respective pathogen and analysed for polyphenol content. The results shows that polyphenol content of diseased leaves is higher than the normal plants. From 7 days old plants as well as from 15 days old plants of sesamum caffeic acid, cinnamic acid, sinapic acid and p- coumaric acid were identified using thin layer chromatography. Their presence was also confirmed by HPLC or co-chromatography with reference compounds and quantified on the basis of integration areas of the peaks of the standard compounds of known concentrations. The results show that while there are a total of 7 major peaks in the control plants, this number is increased to 8 in the plants exposed to pathogen. Besides there are differences in minor peaks further, it is also observed that the amount of total phenolics in the inoculated plants is 25% higher than the controls. On the basis of co - chromatography with the authentic samples, four of these peaks in both the control and inoculated plants were identified as of caffeic acid, hydroxycinnamic acid, sinapic acid and p- coumaric. It is further noted that caffeic acid shows maximum increase on infection by pathogen. It is concluded that exposure of plants with the pathogen indicates not only in the increase of the amount of total polyphenol, but a few additional compounds are synthesized.
