Tonneau 1 is essential for root organogenesis. Lupinus albus (L. albus) forms cluster roots in response to P–deficiency. EST sequencing from preemergent cluster roots 7 days after P – stress identified a Tonneau 1 (LaTON1) gene. The gene is expressed on similar level as actin and alfa-tubulin. LaTON1 cDNA is 66% identical to A. thaliana TON1B and the gene is composed of 8 exons – one more than A. thaliana gene. The two distinct TON1 cDNAs with 3 amino acids indel within the 3’ end region of coding sequence cloned by combined RACE and RT PCR approach represent either two genes or two alleles and code for 256 – 268 amino acids peptides. L. albus TON1 sequences – gene and two cDNA clones are of 95 - 97% identity on nucleotide level and code for peptide with LIS1 motif in the N-terminal part of protein. The proteins with LIS1 motif mediate dynein activity and regulate microtubule dynamics in plants and animals. The gene function was studied with RNAi technique using pHellsGate based construct comprising 3’ end part of LaTON1 coding sequence and 3’ end UTR. Transformation of L. albus and Medicago truncatula roots with a LaTON1 RNAi construct resulted in impaired root development. Roots were stunted, thick and the root meristem was disorganized. TON1 gene function in roots organogenesis is linked to the microtubule dynamics and spatial control of cells division.
