Plant-Pathogen Interactions Griffiths, Jonathan S. [1], Wang, Aiming [2]. Isolation and Characterization of Arabidopsis Translation Initiation Factors Required in Plum Pox Virus Infection. The replication of single stranded positive sense RNA viruses in infected cells is initiated by the synthesis of viral proteins that are required for the replication of viral RNA. Viruses encode a limited number of proteins with well defined functions, and depend primarily on host factors for translation and replication. Identification and removal of host translation factors vital to viral translation, yet dispensable to host biology can lead to developing resistance to viral diseases. To identify candidate genes of host factors, differentially regulated translation initiation factors in Arabidopsis in response to Plum pox virus (PPV) infection were screened at the full genome level using microarray analysis with Affymetrix ATHI GeneChips. There are approximately 124 known translation factors in Arabidopsis and of these, 52 were up-regulated >2.0 fold, and of these four were selected for functional analysis. Salk T-DNA insertion lines for these 4 differentially regulated translation factors were screened for homozygous lines and then challenged with PPV infection. Preliminary results indicate that one of them is required for PPV infection. Currently, protoplasts from Salk insertion lines of this gene were isolated and transfected with a PPV infectious clone to determine if this gene is required for viral replication at a single cell level. Identification of translation initiation factors as host factors is vital to a better understanding of viral infection mechanisms and the development of novel strategies to control PPV.
