We have developed a set of 25 microsatellites for the cycad Cycas micronesica from an EST library from the related species, C. rumphii, and optimized them to describe its population structure across the Mariana Islands using Bayesian approaches. Cycas micronesica, endemic to the Mariana Islands, is of evolutionary, ecological, ornamental, and of medical importance, as it has been implicated in the Alzheimer-Parkinson disease complex among the Chamorro people of Guam. Wild C. micronesica populations in Guam are currently threatened by an invasive aphid, accidentally introduced in 2003 and rapidly spreading, thus far killing all of the saplings and most of the adult plants throughout their distribution. Here, we show the population genetic patterns of a small ex situ collection (NYBG and FBG) and herbaria specimens as preliminary results. Next, we will describe gene flow within and among populations within Guam, focusing on identifying genetic conservation priorities. We we also clarify the biogeographic and taxonomic relationships of C. micronesica within the larger C. rumphii complex in the South Pacific and Micronesia adding LEAFY and atp-rbcL markers. This study will provide the framework for practical conservation based on the levels of genetic variation and gene flow, and evolutionary insights of the biogeographic history of the species complex in the region.
