The use of protease inhibitors to induce resistance of plants to insect pests. Insect pests are one of the serious problems that can cause great economical loss during pea cultivation. Integrated plant protection is a modern trend against pest and diseases, and includes also creation of resistant varieties. This goal can be achieved utilizing biotechnological tools, especially transgenosis. For production of pea plants resistant to insect pests the strategy of expression of an insect protease inhibitor driven by a constitutive promoter was used. The protease inhibitor GmSPI2 was isolated by Nirmala et al. (2001: Eur. J. Biochem., 268: 2064-2073) from Galleria mellonella. A construct pWell09 containing 35S promoter, GmSPI2, and OCS terminator was prepared in the binary vector pGreenII (John Innnes Center, UK). Pea plants were transformed by the transformation method of Svabova et al. (Biol. Plant., 49: 361-370, 2005). Transgenic lines have been prepared for testing of transgene expression and insect resistance. This work was supported by a grant of the Ministry of education CR 1M06030.
